Presentation Title: Target Identification for Muscle Endogenous Repair Using a Novel High-Throughput 3D Model
Abstract: Muscle endogenous repair is the process by which muscle stem cells regenerate muscle tissue in response to injuries. Muscle regeneration fails in aged individuals or in individuals with dystrophies, therefore identifying drugs that can improve muscle regeneration would be highly impactful. Muscle stem cells’ functionalities cannot be characterized on 2D surfaces that are commonly used for in vitro screening; thus, discovering drugs that would improve muscle regeneration has not had much success. McGuigan lab and Gilbert lab have jointly created an engineered 3D tissue model -named MEndR- capable of recapitulating both proliferation and differentiation of muscle stem cells, and therefore Muscle Endogenous Repair. This model has shown great potential in predicting in vivo drug responses in vitro, however the throughput of MEndR is currently low. Increasing the throughput of this system will allow for removal of the guesswork required to identify targets as it will facilitate testing for several potential targets in vitro rather than performing lengthy in vivo experiments. This project has taken steps in creating a novel 96-well platform that models muscle endogenous repair. Once this step is complete, it will be used to identify targets that could improve muscle endogenous repair.
