Presentation Title: Development of an injectable thermoresponsive adhesive embedded with a nanoparticle system for local delivery of a non-steroidal anti-inflammatory drug.
Osteoarthritis (OA) is the most common form of arthritis with well-understood pathologies, resulting in pain and inflammation to the joints. The field’s focus on therapeutic interventions have included the use of non-steroidal anti-inflammatory drugs (NSAIDs) to alleviate these symptoms, one example is celecoxib (CXB), a COX-2 inhibitor typically taken as an oral tablet. However, prolonged systemic intake of CXB can have secondary effects on organs. Hence, the field has moved towards investigating localized delivery of therapies, with intra-articular drug delivery receiving increasing attention. In such systems, the drug can be immediately injected to the site of inflammation to provide instantaneous relief and thereby minimizing systemic effects. A challenging limitation of this latter approach has been short retention time to the site of injection, thereby requiring frequent injections. To overcome this disadvantage, the research thesis presented here is investigating a novel adhesive thermoresponsive hydrogel containing self-assembled nanoparticles loaded with an NSAID. As a first model drug to demonstrate the proof of concept, CXB will be used for intra-articular injection into the knee of an OA patient. The sustained release over a prolonged period will be modeled in vitro and biocompatibility assays will be conducted on the material with cartilage cells, i.e. chondrocytes.
