Presentation Title: A Functional Biomarker of ICUAW to Detect Patient Outcomes in Cell Culture
Abstract: The skeletal muscle comprises a large portion of our body mass and is essential for a variety of body functions. As a result, conditions that affect other body systems can promote skeletal muscle atrophy and weakness. Intensive care unit acquired weakness (ICUAW) is a common complication where 32-50% of intensive care unit (ICU) patients develop muscle atrophy, promoting long-term disability and increasing risk of mortality. The diagnostic limitations and uncertainty as to what factors regulate ICUAW curb its early recognition. Early mobilization is the standard treatment used to ameliorate muscle atrophy and weakness; however, this is a costly and labour-intensive strategy with heterogenous outcomes. We posit that the lack of effective diagnosis and treatment options for ICUAW represents a significant unmet need. To address this problem, our lab has successfully developed a cell culture platform that supports the formation and maturation of 3D human skeletal muscle microtissues (hMMTs). Manifestations of ICUAW including reduced muscle mass, strength, and irregular calcium handling, can be assessed by including ICU patient serum into the hMMT culture media. Successful modelling of ICUAW will allow for further investigation of its etiology.
